
Skin longevity is attracting growing scientific and commercial interest. Retinoids, antioxidants, collagen peptides, hyaluronic acid, hormonal therapies and senescence-targeting agents are increasingly presented as strategies to prevent-or even reverse-the visible signs of skin aging.
But how strong is the clinical evidence?
A new review published in Biochemical Pharmacology by Anna-Lena Gürtler and colleagues critically evaluates the principal topical and systemic interventions currently proposed for healthy skin aging. The authors examine not only their biological mechanisms, but also essential pharmaceutical questions: Are the active ingredients chemically stable? Can they penetrate the skin or be absorbed orally? Do they reach their biological targets? And are their effects supported by reliable clinical measurements?
The review identifies a substantial gap between commercial claims and robust clinical evidence.
Among the available interventions, retinoids remain the most consistently supported, although irritation and limited tolerability can restrict their use. Evidence for antioxidants, hormonal therapies, hyaluronic acid, collagen peptides and several other widely promoted approaches remains heterogeneous or inconclusive.
The authors also highlight recurrent methodological limitations: small study populations, short intervention periods, subjective outcome measures, inadequate control groups and frequent industry sponsorship. These weaknesses make many reported benefits difficult to validate or generalize.
The message is not that skin aging cannot be modulated. Rather, the field must move beyond attractive claims toward interventions supported by validated mechanisms, optimized delivery, objective biomarkers and independent, well-controlled clinical trials.
