Making Skin Ageing Measurable: Epigenetic Age Prediction from Non-Invasive Sampling

Angel Menendez Vazquez, Mitra Bio, United Kingdom

We are pleased to announce Dr. Angel Menendez Vazquez from Mitra Bio, United Kingdom, will join the meeting and will present a talk about Making Skin Ageing Measurable: Epigenetic Age Prediction from Non-Invasive Sampling.

Skin is both the most visible and most environmentally exposed organ, with apparent aging phenotypes. DNA methylation clocks faithfully capture the progression of aging, but so far have been limited to training on abundant in vitro material or invasively collected samples. Here, we demonstrate that skin biological age can be measured directly from a person’s face with superior accuracy, using non-invasive tape-stripping, enabling longitudinal analyses. This model, MitraClock, was validated on independent, longitudinal, and external datasets and was compared against established clocks. It predicts age accurately, with an error of approximately 4 years, outperforming others on epidermal samples. It maintains high accuracy at low sequencing depths, enabling cost-effective scalability and shows intra-individual prediction variation <2 years, highlighting its reproducibility. Its predictive capacity generalised across anatomical sites, conversion and sampling methodologies and on in vitro material. It also successfully captured the rejuvenating effects of Yamanaka factor treatment.
